Preparation of Solutions
« Reply #2 on: November 24, 2012, 09:28:44 AM »
Preparation of Solutions

A. Calculation of Molar, % and "X" Solutions .

1. A molar solution is one in which 1 liter of solution contains the number of grams equal to its molecular weight. Ex. To make up 100 ml of a 5M NaCl solution = 58.456 (mw of NaCl) g/mol x 5 moles/liter x 0.1 liter = 29.29 g in 100 ml of solution

2. Percent solutions.Percentage (w/v) = weight (g) in 100 ml of solution; Percentage (v/v) = volume (ml) in 100 ml of solution. Ex. To make a 0.7% solution of agarose in TBE buffer, weight 0.7 of agarose and bring up volume to 100 ml with TBE buffer.

3. "X" Solutions. Many enzyme buffers are prepared as concentrated solutions, e.g. 5X or 10X (five or ten times the concentration of the working solution) and are then diluted such that the final concentration of the buffer in the reaction is 1X. Ex. To set up a restriction digestion in 25 μ l, one would add 2.5 μ l of a 10X buffer, the other reaction components, and water to a final volume of 25 μ l.

B. Preparation of Working Solutions from Concentrated Stock Solutions

Many buffers in molecular biology require the same components but often in varying concentrations. To avoid having to make every buffer from scratch, it is useful to prepare several concentrated stock solutions and dilute as needed. Ex. To make 100 ml of TE buffer (10 mM Tris, 1 mM EDTA), combine 1 ml of a 1 M Tris solution and 0.2 ml of 0.5 M EDTA and 98.8 ml sterile water. The following is useful for calculating amounts of stock solution needed: C i x V i = C f x V f , where C i = initial concentration, or conc of stock solution; V i = initial vol, or amount of stock solution needed C f = final concentration, or conc of desired solution; V f = final vol, or volume of desired solution

C. Steps in Solution Preparation:

Refer to a laboratory reference manual for any specific instructions on preparation of the particular solution and the bottle label for any specific precautions in handling the chemical. Weigh out the desired amount of chemical(s). Use an analytical balance if the amount is less than 0.1 g. Place chemical(s) into appropriate size beaker with a stir bar. Add less than the required amount of water. Prepare all solutions with double distilled water When the chemical is dissolved, transfer to a graduated cylinder and add the required amount of distilled water to achieve the final volume. An exception is in preparing solutions containing agar or agarose. Weigh the agar or agarose directly into the final vessel. If the solution needs to be at a specific pH, check the pH meter with fresh buffer solutions and follow instructions for using a pH meter. Autoclave, if possible, at 121 deg C for 20 min. Some solutions cannot be autoclaved, for example, SDS. These should be filter sterilized through a 0.22 μ m or 0.45 μ m filter. Media for bacterial cultures must be autoclaved the same day it is prepared, preferably within an hour or two. Store at room temperature and check for contamination prior to use by holding the bottle at eye level and gently swirling it Solid media for bacterial plates can be prepared in advance, autoclaved, and stored in a bottle. When needed, the agar can be melted in a microwave, any additional components, e.g. antibiotics, can be added and the plates can then be poured.

Concentrated solutions, e.g. 1M Tris-HCl pH=8.0, 5M NaCl, can be used to make working stocks by adding autoclaved double-distilled water in a sterile vessel to the appropriate amount of the concentrated solution.

D. Glassware and Plastic Ware

Glass and plastic ware used for molecular biology must be scrupulously clean. Dirty test tubes, bacterial contamination and traces of detergent can inhibit reactions or degrade nucleic acid.

Glassware should be rinsed with distilled water and autoclaved or baked at 150 degrees C for 1 hour. For experiments with RNA, glassware and solutions are treated with diethyl-pyrocarbonate to inhibit RNases which can be resistant to autoclaving. Plastic ware such as pipets and culture tubes are often supplied sterile. Tubes made of polypropylene are turbid and are resistant to many chemicals, like phenol and chloroform; polycarbonate or polystyrene tubes are clear and not resistant to many chemicals. Make sure that the tubes you are using are resistant to the chemicals used in your experiment. Micro pipet tips and microfuge tubes should be autoclaved before use.

Rafa looks comfortable on first day at Chelsea
« Reply #3 on: November 24, 2012, 06:32:16 PM »
Nov 24, 2012

In many ways, it was as if Rafa Benitez had never been out of the game. For a good 10-minute stretch during his first Friday press conference at Cobham, the new Chelsea manager once again had the bottles and glasses arranged on the table as he attempted to explain the intricacies of zonal marking.
It wasn't the only Benitez hallmark. There was also a barb in response to Alex Ferguson, as he spoke of how he saw Arsene Wenger as the best manager in England when he first arrived in the country. If it was a throwaway line, it was definitely in a deliberate direction.

In truth, though, the talk about mindgames was the only moment when the press conference got any way tetchy, if even that. For the most part, Benitez was in as good a mood as he can be. It was also then, though, that you realised just how happy he was to be back in the game.

"It's something I've been waiting for. I've been working, writing books, websites, analysing games, but I like to be on the pitch. When I came here from Valencia, I said I liked to be on the pitch, communicating with players, giving them my ideas. The problem is I don't have much time. I'm really pleased to be on the pitch talking about football."
Benitez also admitted he had a good chat with the senior players, but didn't need to build any bridges.

4th International Conference on Education & Educational Psychology

2-5 October 2013

Antalya, Turkey

website :http://www.iceepsy.org/

I'm deeply honoured to chair the 4th International Conference on Education and Educational Psychology and I am delighted to invite you to attend. Students, researchers, practitioners, educators, teachers, psychologists, educationalists, we all have this great opportunity to come together at the beautiful Antalya by the Mediterranean coast of Turkey to share cutting-edge thinking and together find effective solutions to the key challenges that we are facing.

Important challenges come into local or larger guises. There are many local challenges that can be overcome through international collaboration. We live in a world of increased international mobility; not only we as professionals are more mobile, but also the people to whom we provide services come from a range of countries and cultural backgrounds. We need to be responsive to such diversity, whilst utilising it as a source of strength and as an opportunity for innovation and improvement.

ICEEPSY 2013 has every potential to build on the successes of its three predecessors. ICEEPSY 2011 has attracted over 370 presentations from 45 countries, and ICEEPSY 2012 has advanced even that with over 450 registered participants from 57 countries. The ICEEPSY 2013 Scientific Committee and Board of Reviewers are working hard to ensure that the quality of this event exceeds any precedent.

In the established tradition of the previous three Conferences, ICEEPSY 2013 will publish all accepted full-text papers in Procedia - Social and Behavioral Journal (ISSN: 1877-0428 - ELSEVIER) and indexed in ScienceDirect, Scopus & Thomson Reuters Conference Proceedings Citation Index (ISI, Web of Science). Moreover, accepted abstracts will be published in the Abstract Book ICEEPSY (ISSN: 1986-3020), a serial publication that has been registered with the International Centre for the Registration of Serial publications in Paris.

ICEEPSY 2013 especially welcomes papers from the following and related topics: Assessment and Evaluation, Teacher Education, Basic Education, Basic Skills; Counselling Psychology; Adolescent Growth and Development, Educational Psychology; Continuing Education, Higher Education, Quality Assurance / Institutional Effectiveness; Learning Theories, Effective Teaching Practices, Second Language Teaching; New approaches in Psychology; Special Education, Education and Technology, ICT, Distance Learning.

The quality of ICEEPSY 2013 is further accentuated by its internationally renowned keynote speakers. ICEEPSY 2013 will, in addition, feature a range of interesting workshops as well as a Special International Roundtable for participants to engage in additional discussions related to the key topics addressed at the Conference and further cross-fertilisation across thematic sections.

I am also delighted that the organisers of the Conference are working hard to ensure that the scientific quality of the Conference is mirrored by the practical aspects of our stay. I am looking forward to discovering Antalya’s Mediterranean Gulf, its surrounding mountains and forests, and the ancient ruins, as well as enjoying the renowned local cuisine.

I look forward to seeing you in Antalya in October 2013.

Some highest International Bridge............
« Reply #6 on: November 27, 2012, 10:59:57 PM »
1. Siduhe River Bridge. Yesanguanzhen, Hubei, China.

Description: 1,627 feet high / 496 meters high
2,952 foot span / 900 meter span.

With a roadway 1,627 feet (496 mtrs) above the water, the Siduhe River bridge is the latest Chinese champ to take the record as the highest bridge in the world. Opened on November 15th, 2009, it is the 3rd Chinese bridge in less than a decade to claim the title of “World’s Highest Bridge” and is a symbol of just how fast and how far China’s highway infrastructure has come in such a short period of time.

Located about 50 miles (80 km) south of the famous 3 Gorges region of the Yangtze River in China’s mountainous Hubei Province, the Siduhe suspension bridge is just one of several amazing structures on the last 300 mile (483 km) link of the 1,350 mile (2,175 km) long West Hurong highway that now connects Shanghai on the Pacific coast with the cities of Chongqing and Chengdu in the west.

The 8 highest of China’s many high bridges were all built after 2000 and are located primarily in the 3 western Provinces of Guizhou, Hubei and Chongqing. All 8 of these bridges exceed 900 feet (274 meters) in height and rank among the world’s 12 highest while four have held the world record for highest road or rail bridge. Even more amazing, 6 of these bridges are higher than Colorado’s Royal Gorge, the former world’s highest bridge from 1929 to 2001.

2. Baluarte Bridge. El Palmito, Sinaloa, Mexico
Description: 1,321 feet high / 403 meters high
1,706 foot span / 520 meter span

The Baluarte River Bridge is not only the highest bridge in North America but the highest cable stayed bridge in the world surpassing the Millau Viaduct in France. It is the crown jewel of the greatest bridge and tunnel highway project ever undertaken in North America. Known as the Durango-Mazatlán highway, it will be the only crossing for more than 500 miles (800 km) between the pacific coast and the interior of Mexico. The path of this new highway roughly parallels the famous “Devil’s Backbone”, a narrow road that earned its nickname from the way it follows the precarious ridge crest of the jagged peaks of the Sierra Madre Occidental mountains. The dangerous road is a seemingly endless onslaught of twisting, terrifying turns that are so tight there are times the road nearly spirals back into itself.

3. New River Gorge Bridge.. Fayetteville, West Virginia, United States

Description: 876 feet high / 267 meters high
1,700 foot span / 518 meter span, 1977

A monumental steel arch built on a grand scale not often seen in bridge construction, the New River Gorge bridge in the U.S. state of West Virginia opened in 1977 as the highest and longest arch bridge in the world with a height of 876 feet (267 mtrs) and a main span of 1,700 feet (518 mtrs). Several Chinese bridges eroded these records away, first in 2001 when the Beipanjiang railway arch bridge opened with a greater height of 902 feet (275 mtrs) and then in 2003 when the Lupu arch bridge opened in Shanghai with a longer main span of 1,804 feet (550 mtrs). Then in 2009, China finally opened an even higher 4-lane road arch in the form of the Zhijinghe bridge with a deck 965 feet (294 mtrs) above the water - 90 feet (27 mtrs) higher then the West Virginia span.

Built at a cost of 37 million dollars, the bridge was designed by the large engineering firm of Michael Baker, Jr. and constructed by the legendary American Bridge Company. The massive span itself was built by using a highline with two 330 foot (101 mtr) tall towers some 3,500 feet (1,067 mtrs) apart.

4. Italia Viaduct.. Laino Borgo, Calabria, Italy
Description: 850 feet high / 259 meters high
575 foot span / 175 meter span,1974

The highest bridge in Italy, the Italia viaduct was the second highest bridge in the world upon its opening in 1974. It is still the highest steel beam bridge in the world and was the highest bridge in Europe for 30 years until France’s Millau Viaduct surpassed it in 2004. Only China’s concrete Liuguanghe is higher among all beam bridges. The viaduct is the highest among several impressive bridges along Italy’s spectacular Autostrada Napoli-Reggio Calabria or A3 motorway including the biggest and highest frame bridge in the world over Sfalassa gorge.

Soaring 850 feet (259 mtrs) above a deep cut over the Lao river, the Italia Viaduct lives up to its name with a very Italian-style steel box beam design. The central portion of the bridge consists of a continuous single beam of 1,394 feet (425 mtrs) spread over 3 spans including a central river span of 575 feet (175 mtrs). The 472 foot (144 meter) tall pier is Italy’s fourth highest. The Gorsexio and Sente bridges have higher piers of 541 feet (165 meters) as well as the Rago Viaduct at 482 feet (147 meters).

5. Phil G McDonald Bridge,Glade Creek Bridge.. Beckley, West Virginia, United States

Description: 700 feet high / 213 meters high
784 foot span / 239 meter span,1988.

.

Of North America’s 10 highest bridges, the Phil G McDonald or Glade Creek bridge is probably the least known. Located several miles east of Beckley, West Virginia, U.S.A., this huge deck truss has one of the tallest piers ever built on an American bridge, rising 341 feet (104 mtrs) from the foundation to the underside of the truss. The shorter pier measures 293 feet (89 mtrs) tall.

Coincidentally, Glade Creek is a near twin of California’s Auburn-Foresthill bridge. From an engineering standpoint, the only major difference between the two is the West Virginia span is a continuous truss instead of a cantilever. The California span is only about 10 percent bigger in size and height. Much of Glade Creek’s status as a “hidden” high bridge is probably due to its being in the shadow of the world famous New River Gorge bridge located just 20 miles (32 km) to the north. Even so, at 700 feet (213 mtrs), Glade Creek is exactly 80 percent as high as the New River span. And although it does not have the same prestige as “The World’s Highest Arch Bridge”, Glade Creek still holds a record of its own as the “World’s Highest Truss Bridge”.

The thick foliage surrounding the structure make it a difficult bridge to photograph. A graded dirt road on the west side of the bridge leads down into the ravine where you can hike along the creek and look up at the deck, 70 stories above.

6. Kochertal Viaduct.. Braunsbach, Baden-Württemberg, Germany.
Description: 607 feet high / 185 meters high
453 foot span / 138 meter span,1979

he highest bridge ever built in Germany, the massive Kochertal Viaduct was the tallest viaduct in the world for 25 years until 2004 when France’s Millau Viaduct finally surpassed it. China also opened two taller viaducts in 2008 and 2009 in the form of the Mashuihe and Longtanhe bridges on the last section of the West Hurong highway between Shanghai and Chengdu. Although there is no absolute definition of what constitutes a viaduct, such bridges usually cross wide valleys with several spans of equal length instead of one large central span. Kochertal’s tallest pier is tied with Longtanhe as the 5th highest in the world measuring 584 feet (178 mtrs). Only the Millau Viaduct and Mexico’s El Carrizo and San Marcos bridges and China's Labajin have taller piers.

Opened in 1979, the Kochertal carries Germany’s A6 motorway 607 feet (185 meters) above the broad Kocher River valley. Constructed outward from both sides of the valley using formworks, the massive 21 foot (6.5 meter) deep single cell box girder was launched first before inclined struts were placed at 25 foot (7.66 meter) intervals from the bottom edge, allowing the wider wings of the road deck to cantilever out to the side from the central box beam. The total width of the massive deck is 102 feet (31 meters). The clean lines of the bridge come from architect Hans Kammerer as well as the engineers at Leonhardt, Andrä und Partner and Dycherhoff & Widmann.

Country-level situation analysis of nutrition security in African countries
« Reply #10 on: November 28, 2012, 12:45:40 PM »
Malawi:
Sustained economic growth proving elusive. High proportion of population unable to
meet basic needs. High levels of child malnutrition. Very low purchasing power results in a poor,
unreliable market system. Potentially productive agricultural sector, but subject to floods and
drought, so aggregate food availability not assured. Landlocked country, so costs of imports,
including agricultural inputs, are high, and exports may be uncompetitive globally owing to high
transport costs. Difficult to make profitable use of high-yielding agricultural technologies, resulting
in unsustainable mining of natural resources. Emergency food management system proved deficient in last emergency (2002–03). HIV infection relatively high.

Mozambique:
Encouraging signs for sustained economic growth, but high proportion of population unable to meet basic needs. Potentially productive agricultural sector but subject to cyclones, floods, and drought. Considered relatively food insecure. Poor marketing and transport systems. Sparse health infrastructure in rural areas, with high levels of child mortality. HIV infection relatively high. Low female literacy, but girls increasingly are going to school. Limited numbers of trained professionals in agriculture and nutrition sectors, particularly outside the capital. Reliant on international NGOs to deliver many social services in rural areas. Potentially good access to global trade. Food security and nutrition relatively important policy issues.

Nigeria:

Major global oil producer. Economic diversification away from oil not yet successful. Oil benefits not equitably distributed across population. Sustainable, broad-based economic development proving difficult to achieve. Diverse and potentially productive agricultural sector. High proportion of population reliant on agriculture. Food secure in terms of aggregate food availability. However, many unable to meet basic needs, including in food. High levels of malnutrition in spite of availability of food. Dynamic but crumbling market system. Good linkages to global market systems. Policy processes are disordered, unpredictable, and not transparent, with little effective master development planning. However, recently passed a national policy on food and nutrition. Large numbers of trained professionals in agriculture and nutrition, but ineffectively utilized. Health and educational systems in place, but quality of performance is quite varied. Sharp contrasts in educational attainment levels across the country for both men and women. In contrast to the other countries, level of donor support quite low on a per capita basis.

Uganda:

Encouraging economic growth and poverty reduction over the past decade. Relatively effective and transparent policy processes. Strong emphasis on decentralized, democratic decisionmaking,
although central government remains dominant in policymaking and resource allocations.
Nevertheless, armed conflict in the north and northeast is causing significant food insecurity.
Agriculture is focus of economic growth strategies. Relatively productive agriculture, with
sufficient food available in aggregate. Landlocked nation, so comparative economic advantage
within global markets is problematic. Education and health systems are in place, if somewhat
overstretched. Limited numbers of professionals, but used relatively effectively.

« Reply #13 on: November 29, 2012, 09:44:55 AM »

Mix masters: NIST scientists image the molecular structure of polymer blends

Typical BCARS composite image of a polyethylene blend taken at NIST showing circular polarization response. LLD polyethylene shows red in this mode, while the HD polyethylene with deuterium substituted for hydrogen is green.
NIST

Using an enhanced form of "chemical microscopy" developed at the National Institute of Standards and Technology (NIST), researchers there have shown that they can peer into the structure of blended polymers, resolving details of the molecular arrangement at sub-micrometer levels. The capability has important implications for the design of industrially important polymers like the polyethylene blends used to repair aging waterlines. Polyethylene is one of the most widely produced and used polymers in the world. It's used in many familiar applications -- milk bottles, for instance -- but the NIST research is motivated by a more critical application: water pipes. Aging water infrastructure is a significant national issue. The Environmental Protection Agency has reported that in the United States there are over 240,000 water main breaks per year, leaks wasting 1.7 trillion gallons of water per year, and costs to taxpayers of $2.6 billion per year.
