Recombinant production of functional full-length and truncated human TRAM/TICAM-2 adaptor protein involved in Toll-like receptor and interferon signaling


Highlights

 Full-length and truncated TRAM proteins were successfully expressed in E. coli and purified.
Systematic construct design and small-scale expression helped to determine the boundaries of the TRAM TIR domain.
 TRAM and TLR4 TIR domains interacts physically; the BB loop regions of both proteins are crucial for this interaction.
 Large-scale purification and biophysical characterization identified a TRAM construct suitable for structural studies.
